Extraction of Se-polysaccharides from Taxus chinensis by orthogonal test

Abstract
In this thesis, Taxus chinensis, a kind of precious medicinal plant that grows in Enshi where it is a high selenium areas in China, was used as experimental material. Solid -liquid ratio, water extraction time and microwave processing time were the three experimental factors, then orthogonal experiment was designed to find a more efficient way to extract and separate Se-polysaccharide from T. chinensis. Based on that, the phenol sulfuric acid method was used to detect the content of Se-polysaccharide in T. chinensis, and the atomic fluorescence method was used to detect the content of Se in Se -polysaccharide. In addition, statistical software, SPSS, was used to analyze and process the orthogonal experiment results. The results showed that the best conditions to extract this kind of polysaccharide were as follows: the solid-liquid ratio was 180 (W/V), water extraction time was 130 min, microwave processing time was 5 min. Among these three factors, the influence order on polysaccharide extraction rate was solid-liquid ratio>microwave processing time>water extraction time. Under these conditions, the content of Se-polysaccharide was up to 1.96%, selenium content in Sepolysaccharide was 0.0555 g/g.
