    Isolation and identification of endosymbiont from brown planthopper, Nilaparvata lugens (Stl)

    • To better understand the endosymbionts description in brown planthopper (BPH), Nilaparvata lugens (Stl), Gause's Synthetic Agar and potato dextrose agar (PDA) were used to culture the intestine of BPH, and endosymbiont strains were isolated and purified, then the 16S rDNA of these strains were amplified by PCR with the universal primers. The sequence was aligned by Blast tool, and showed 99% of similarity with Arsenophonus sp. of Aleyrodidae, which indicated that the strain obtained belonged to Arsenophonus. To construct the phylogenetic tree by Neighbour-Joining method, 7 sequences of 16S rDNA from endosymbionts of Aleyrodidae were adopted, and it showed a closer relationship with a sequence GU124505 from BPH, which may be the same species with the strain purified.
