Study on callus induction and cell suspension culture of Morinda citrifolia

Abstract
Faint yellow and loose callus was induced from vein of Morinda citrifolia on B5 medium. After proliferation, the callus was inoculated in liquid medium and cell suspension lines were established. Medium variety, inoculation densities, pH and sucrose concentration were optimized in this research. The results indicated that, faint yellow and loose callus could be induced on B5 medium by adding 1 mg/L 2,4-D and 0.1 mg/L 6-BA; B5 medium by adding 2 mg/L NAA and 0.2 mg/L KT was the better callus proliferation medium. Cells could grow well with 2 mg/L NAA and 0.2 mg/L KT in B5 medium with 60 g/L (fresh cell weight) of inoculation densities, 4% sucrose, pH 5.8, 100 r/min, and 27~28益in dark. The specific growth rate was 0.0836 d-1, and anthraquinone production could reach 367.8 mg/L after 27~30 d of suspension culture.
