Effects of overexpression of H3K9me3 demethylase on the in vitro developmental efficiency of cloned porcine embryos

Abstract
To explore the effects of overexpression of histone H3K9me3 demethylases KDM4B/KDM4D on the in vitro developmental efficiency of porcine cloned embryos,KDM4B/KDM4D expression vectors were constructed, and KDM4B/KDM4D mRNA were obtained by in vitro transcription and were microinjected into cloned pig embryos, respectively. The results show that injected cloned embryos have a similar H3K9me3 level,cleavage rate and blastocyst rate with non-injected cloned embryos,but have a significantly higher KDM4B/KDM4D expression level than non-injected control cloned embryo( P<0.05),and cloned embryos injected with KDM4B mRNA have a significantly higher total number of cells per blastocyst than non-injected control cloned embryos( P<0.05). These results suggest that overexpression of histone H3K9me3 demethylases KDM4B can significantly improve the in vitro developmentral rate of cloned porcine embryos.
