    Optimization of high-frequncy regeneration system of Alfalfa

    • Hypocotyls of 3 varieties of Alfalfa in 7 days old seedlings were used as explants,Ms medium was used as basic medium,the effects of different concentrations of auxins on Alfalfa callus regeneration system,adventitious bud induction and rooting were studied. The results indicated that, for Golden Empress,Gan-nong No.3 and Algonquin,the best mediums for callus induction were MS+2,4-D 2.5 mg/L+6-BA 0.5 mg/L,MS+2,4-D 2.0 mg/L+NAA 1.0 mg/L,MS+2,4-D 3.0 mg/L+KT 1.0 mg/L;the best differentiation mediums were MS+6-BA 2.0 mg/L+CH 0.5 g/L,MS+NAA 0.3 mg/L+KT 0.5 mg/L,MS+KT 1.2 mg/L; the best rooting medium for plant development were 1/2MS+IBA 1.0 mg/L.
