甜瓜 ERF 转录因子基因 CmERFII-11cDNA 克隆及表达分析

    Cloning and expression analysis of ERF transcription factor gene CmERFII-11 cDNA in melon(Cucumis melo L.)

    • 摘要: 根据已报道的甜瓜 CmERFII-11 基因 cDNA 序列设计特异性引物, 采用 RT-PCR 技术从甜瓜果实中克隆得到 CmERFII-11 基因。 采用相关分析软件对该基因进行了生物信息学分析, 利用荧光实时定量 PCR进行表达分析。 结果表明, 所克隆的 cDNA 长度为 954 bp, 编码 255 个氨基酸, 编码蛋白质的分子量为 28.33ku, 理论等电点为 7.62。 该基因在根、 茎、 叶组织以及授粉后不同时期果实中均有表达, 在叶片组织中的表达量最高。

       

      Abstract: The specific primers were designed according to reported cDNA sequences of CmERFII-11 in melon, CmERFII-11 gene cDNA was cloned from fruit of Cucumis melo L. by RT-PCR method. CmERFII-11 gene was analyzed by the tools of bioinformatics, expression pattern was showed by Real-time quantitative PCR. The length of cDNA was 954 bp coding a polypeptide of 255 amino acids with a calculated molecular weight of 28.33 ku and an isoelectric point of 7.62. CmERFII-11 gene expressed in the root, stem, leaf and different developmental stages of fruit after pollination, CmERFII-11 gene expressed highest in leaf.
