实时荧光PCR 法特异性检测棉花曲叶病毒

    Rapid detection of cotton leaf curl virus by real-time PCR

    • 摘要: 根据棉花曲叶病毒(Cotton leaf curl virus,CLCuV)运动蛋白基因序列特征,设计并合成了特异性Taqman 探针和检测引物,建立该病毒的实时荧光 PCR 快速检测方法。 结果表明:该方法可以特异性地从 5 个不同 CLCuV 分离物中扩增到荧光信号,而其他 6 种病毒均无扩增。 建立的方法具有快速、准确、灵敏及简便等特点,可为进出境植物检疫和农业安全生产提供可靠的技术支持。

       

      Abstract: In this study, special Taqman fluorescent probes and primers were designed based on the sequence ofmovement protein gene, a rapid and effective method of cotton leaf curl virus (CLCuV) detection was established basedon real-time fluorescent PCR. The result showed that the method could amplify specifically fluorescence signal from 5different isolates of CLCuV, while the other 6 viruses had no amplification. The method is not only rapid, accurate,convenient, and sensitive, but also can provide well technical support for Entry-Exit Inspection and Quarantine andagricultural safety.
