月季、玫瑰FLA基因家族鉴定及其在雌雄蕊中的表达分析

    Identification of the FLA Gene Family in Rosa chinensis and R. rugosa and Analysis of Its Expression in Pistils and Stamens

      目的 类成束阿拉伯半乳糖蛋白(Fasciclin-like arabinogalactan proteins,FLAs)是阿拉伯半乳糖蛋白(Arabinogalactan-proteins,AGPs)的一个亚类,广泛参与植物的生长发育、有性繁殖、非生物胁迫响应等生物过程。对蔷薇属植物,月季(Rosa chinensis)、玫瑰(R. rugosa)中FLA基因家族进行鉴定分析,有助于了解其在蔷薇属植物生物过程中的作用。
      方法 采用生物信息学方法对月季、玫瑰中的FLA基因家族进行全基因组鉴定,将二者的FLA基因家族进行比较,同时分析FLA基因在月季、玫瑰雌雄蕊中的表达模式。
      结果 在月季和玫瑰中均鉴定出25个FLA基因,不均匀分布在各自的7条染色体上。基因结构分析发现月季和玫瑰中分别有18、21个FLA不含内含子,启动子区域顺式作用元件预测显示光响应元件、茉莉酸甲酯响应元件、生长素相关顺式元件分布较广。全部50个FLAs中33个N端存在信号肽、30个为跨膜蛋白、44个可能定位在细胞膜上,且亲缘关系较近的FLAs二级、三级结构、基序分布均较为相似。系统进化分析结果显示,所有FLAs分为6组,Group Ⅰ成员最多且分枝较多,其他组内均有月季和玫瑰FLAs分布。对FLA基因在月季、玫瑰雌雄蕊中的表达模式分析发现,部分FLA在雌蕊或雄蕊中特异性高表达。
      结论 从月季、玫瑰中均成功鉴定出25个FLA基因,其具有保守性及物种特异性。在雌蕊或雄蕊中部分FLA基因特异性高表达,可能参与月季、玫瑰的育性调控,这为进一步研究其对蔷薇属植物生殖发育的调控提供了参考。

       

      Abstract:
      Objective Fasciclin-like arabinogalactan proteins (FLAs) were a subclass of arabinogalactan- proteins (AGPs). The FLAs were widely involved in biological processes such as plant growth and development, sexual reproduction, and abiotic stress response. Identifying and analyzing the FLA gene family in Rosa plants (R. chinensis and R. rugosa) could help to understand the role of the FLA gene family in the biological processes of Rosa plants.
      Method This study employed bioinformatics to conduct a genome-wide identification of the FLA gene family in R. chinensis and R. rugosa. The FLA gene families of the two species were compared, and the expression patterns of FLA genes in the stamens and pistils of R. chinensis and R. rugosa were analyzed.
      Result Twenty-five FLA genes were identified in both R. chinensis and R. rugosa, unevenly distributed across their respective seven chromosomes. Analysis of the structure of FLA genes in R. chinensis and R. rugosa revealed that 18 RcFLA in R. chinensis and 21 RrFLA in R. rugosa did not contain introns. Analysis of cis-regulatory elements found that light response elements, MeJA response elements, and auxin-related cis-elements were widely distributed. Among all 50 FLA proteins, 33 had signal peptides at their N-termini, 30 were transmembrane protein, and all 44 were likely localized on the cell membrane. The closely related FLA proteins showed strong similarity in their two-dimensional and three-dimensional structures and motif distribution. Phylogenetic analysis divided all FLAs into six groups, with Group Ⅰ having the most members and branches, and other groups containing FLAs from both R. chinensis and R. rugosa. Analysis of the expression patterns of FLA genes in the stamens and pistils of R. chinensis and R. rugosa revealed that some FLA were specifically highly expressed in the stamens or pistils.
      Conclusion A total of 25 FLA genes were identified in both R. chinensis and R. rugosa, indicating that these FLA genes are conserved and also species-specific. Additionally, some FLA genes were specifically highly expressed in the pistil or stamen, and were potentially involved in the fertility regulation of R. chinensis and R. rugosa,  providing a reference for further research on the regulation of reproductive development in Rosa plants.
