云斑尖塘鳢烂身病病原分离鉴定及药敏试验

    Isolation,identification and drug susceptibility of Aeromonas veronii isolated from Oxyeleotris marmoratus

    • 摘要: 从患烂身病的云斑尖塘鳢上的溃疡灶中分离到菌株WSQ-01,该菌株在脑心浸出液琼脂培养平板上生长呈边缘光滑、有光泽的圆形白色菌落。通过染色发现该菌为革兰氏阴性菌,用16S rRNA 通用引物进行PCR 扩增、克隆及测序,将获得的序列在GenBank 数据库中进行Blast 比对分析。结果表明,WSQ-01与维氏气单胞菌的亲缘关系最近,同源性超过99%;动物回归实验证实该菌能引起健康云斑尖塘鳢的烂身和死亡,且能重新分离到该菌株。药敏试验结果表明,四环素、强力霉素、红霉素、氟苯尼考、利福平、头孢他啶、氧氟沙星、左氟沙星和恩诺沙星对该菌的作用较强,该菌对这10 种药物敏感,对氨曲南、链霉素和阿莫西林具有耐药性。

       

      Abstract: A highly pathogenic bacterium WSQ-01 was isolated from the diseased Oxyeleotris marmoratus with typical skin ulcer. The bacteria strain was a gram-negative bacillus whose colony was white,round and smooth edges. The 16S rRNA of the bacteria was amplified,cloned and sequenced. The sequence were Blast in GenBank and downloaded the high homology bacterial sequence,then the sequences were aligned,analyzed and constructed the phylogenetic tree. The similarities of the 16S rRNA gene sequences in the strain WSQ-01 to other Aeromonas veronii were more than 99%. According to the phylogenetic tree,the strain JX-096 clustered together with Plesiomonas shigelloides. Animal experiments confirmed that the bacteria can cause skin ulcer and death on health Oxyeleotris marmoratus and have a strong pathogenic effect. Sensitive test of the bacteria showed that the strain highly sensitive to tetracycline,doxycycline,erythromycin,florfenicol,rifampicin,ceftazidime,ofloxacin,levofloxacin and enrofloxacin but resist to amamicin,streptomycin and amoxicillin.
