猪伪狂犬病毒的分离鉴定及其gE、TK 全基因序列遗传变异分析

    • 摘要: 将分离到的8 种PRV 分离株的gE、TK 基因与国内外的毒株进行核苷酸及氨基酸的序列同源性分析,结果发现,分离毒株的gE、TK 基因与15 株国内外已发表的代表毒株核苷酸及氨基酸的序列同源性分别为97.3%~99.9%、98.7%~99.9%;遗传进化分析结果表明,分离毒株与国内近年分离自不同省份的代表毒株亲缘关系较近,与国外分离的代表毒株亲缘关系较远;而对gE、TK 基因氨基酸主要变异位点分析表明,gE 和TK 基因都有一些位点已经发生突变。结果表明猪PRV 广东部分地区分离毒株已发生变异,为规模化猪场对猪伪狂犬病的净化以及疫苗的选择提供参考。

       

      Abstract: The nucleotide and amino acid sequences homology of the gE and TK genes of the eight isolates with published sequences were analyzed, the results showed that the homology of nucleotide sequences of gE and TK were up to 97.3%-99.9%, 98.7%-99.9%, respectively, while the homology of amino acid sequences of gE and TK were 94.6%-99.8% and 98.1%-99.3%. Genetic evolution analysis indicated that the isolates in our study had a far relationship with the recent strains from other provinces and contries, the gE and TK all had multi-sites mutations in the major domain of amino acid sequence. These results indicated that porcine pseudorabies virus in Guangdong province had mutated much,the results could provide candidates of vaccine against PRV and be helpful to prevent PRV.
