广东夏大豆新品种(系)分子身份证的构建

    Establishment of molecular ID of summer soybean varieties( lines) in Guangdong

    • 摘要: 建立快速简便鉴定广东夏大豆种质资源的方法。对大豆20 个连锁群上的159 对SSR 引物进行筛选,其中48 对引物扩增清晰、稳定,用其检测34 份广东夏大豆材料,将数字化后的等位变异片段用资源特征分析软件ID Analysis 1.0 进行分析。结果表明,48 对多态性引物平均等位变异数为3.88,平均基因多样性指数为0.50,平均引物多态性信息含量(PIC)为0.45,可用于区分广东夏大豆资源。仅需5 对引物(Sat_267、Sat_235、Satt277、Sat_351 和Sat_292)可将全部34 份种质区分开,成功构建了一套广东夏大豆种质的分子身份证。

       

      Abstract: To establish a fast and simple method for identifying Guangdong summer soybean varieties( lines),48 primers showing polymorphic bands were screened from 159 SSR primers covering 20 linkage groups of soybean chromosome,and be used to detected 34 Guangdong summer soybean varieties( lines). The data number ranked from fragment size of allele variation were analyzed by the software ID Analysis 1.0 . The average number of alleles by each primer was 3.88,and the average of gene diversity index and polymorphism information content( PIC) of the 48 SSR loci was 0.50 and 0.45,respectively. These primers could be used to identify the Guangdong summer soybean germplasm.Only using five primers( Sat_267,Sat_235,Satt277,Sat_351 and Sat_292) could identify all 34 materials. A set of molecular ID was established for Guangdong summer soybean varieties( lines).
